چکیده

The evaluation of medium for in vitro maturation of buffalo oocytes has progressed towards more defined conditions for the studies evaluating effect of specific components in the medium. Therefore, aim of the present study was to evaluate the effect of fatty acid free-bovine serum albumin (FAF-BSA) alone or supplemented with epidermal growth factor (EGF) in the in vitro maturation medium on in vitro maturation of oocytes and subsequent embryo development. The cumulus oocyte complexes (COCs) were collected from ovaries (900) of slaughtered animals by aspirating follicles, and incubated in maturation medium for 24 h. Three types of maturation media (MM) supplemented with 1) 10% fetal calf serum (FCS), 2) 6 mg/ml FAFBSA or 3) 6 mg/ml FAFBSA + 10.0 ng/mL epidermal growth factor (EGF) were evaluated. After maturation, Tyrode’s albumin lactate pyruvate (TALP) and synthetic oviductal fluid (SOF) media were used for fertilization and culture environments, respectively. The percentage of COCs with expanded cumulus was higher (P < 0.05) in MM supplemented with FBS and FAF-BSA+EGF compared with that having FAF-BSA alone. No difference (P > 0.05) was observed in meiotic resumption and nuclear maturation rate in all three types of maturation media. Percentage of embryos crossing 8-16 cell stage and reaching up to 32 cell stage (morula) after IVC was lower (P < 0.05) in MM supplemented with FAF-BSA, however with addition of EGF, embryo development was improved (statistically non significant) and was comparable (P>0.05) with development in MM supplemented with FCS. In conclusion, serum free defined system having FAF-BSA+EGF was comparable to the MM containing FCS and can be used to replace serum supplemented media for the in vitro maturation of buffalo oocytes and further embryo development.

Background: This study examined the effect of including one of two growth factors (100 ng/ml IGF-1 or 20 ng/ ml EGF) in combination with one of two antioxidants (50 μM cysteamine or 50 μM beta-mercaptoethanol) in invitro maturation of buffalo oocytes, followed by fertilization and subsequent development of embryos. Materials and Methods: The oocytes were recovered by in vivo ovum pick-up techni...

Effects of cysteamine during in vitro maturation on viability and meiotic competence of vitrified buffalo oocytes

The aims of the present study were to assess the effects of cysteamine as an anti-oxidant on the rate of in vitro maturation (IVM) of buffalo oocytes (experiment 1), and their viability and nuclear status following vitrification (experiment 2). Immature oocytes with compact cumulus cells obtained from the ovaries of slaughtered animals were harvested and then cultured in the maturation medium w...

O-11: Diverse Effects of Polyunsaturated Fatty Acids on Oocyte Maturation and Development In vitro

Background: Polyunsaturated fatty acids (PUFAs) have been shown to influence fertility and endocrinology of reproduction and metabolic activity in many species. In dairy cows, we and others have shown changes in steroid and metabolic hormones and prostaglandins leading to alteration of ovarian activity and uterine function. These can influence fertility by changes in folliculogenesis cyclicity ...

In vitro Maturation and Fertilization of Buffalo Oocytes Cultured in Media Supplemented with Bovine Serum Albumin

The aim of this study was to determine the quality of cumulus-oocyte-complexes (COCs) and the effects of bovine serum albumin (BSA) supplementation on in vitro maturation and fertilization rate of buffalo oocytes. COCs were collected from slaughterhouse buffalo ovaries by aspiration method. Only normal grades COCs were matured for 48 hours in TCM-199 media. Two groups were created: one for the ...

Background: The efficiency of in vitro maturation (IVM) has remained low despite extensive effort to understand the culture conditions. This study investigated the effect of cumulus cells and meiotic inhibitors on oocyte developmental competence in the presence of Alpha Lipoic acid (ALA) Materials and Methods: Mouse germinal vesicle oocytes were considered in cumulus-denuded oocytes (CDOs) and ...
